Pihak pembelaan dalam perbicaraan liwat II membabitkan Datuk Seri Anwar Ibrahim, hari ini menimbulkan kemungkinan sampel yang diambil dan diuji oleh ahli kimia dicemari tetapi hakikat itu diabaikan.Tumpuan adalah terhadap calitan kapas B9 yang diperolehi dari kawasan rektum Mohd Saiful Bukhari Azlan dan tuala putih yang diperolehi daripada sel Anwar apabila beliau ditahan semalaman.
Ram Karpal, yang menyoal balas berkata demikian berikutan penemuan 18 allele yang menunjukkan kemungkinan kehadiran orang ketiga. Katanya, keraguan timbul tentang prosedur standard makmal Jabatan Kimia Malaysia (JKM) bersabit sampel DNA yang diperolehi daripada pengadu Saiful Bukhari.
Pakar DNA Australia Dr Brian McDonald berkata kedua-dua ahli kimia tempatan Dr Seah Lay Hong dan Nor Aidora Saedon - tidak sepatutnya mengabaikan pencemaran berkenaan.
"Mereka sepatutnya menjalankan ujian lanjut untuk memperkuatkan penemuan tentang 18 allele itu," kata McDonald.
Katanya, berdasarkan laporan pakar kimia tempatan itu, mereka telah menemui tiga sampel DNA dari dubur Saiful.
Namun Dr McDonald berkata Seah tidak menyatakan dalam laporannya akan kehadiran DNA individu ketiga yang menyumbangkan bacaan tinggi melalui ujiannya selain daripada Saiful dan 'lelaki Y'.
"Dia (Seah) menemui DNA lain tapi dia tidak melakukan apa-apa," katanya.
Apabila ditanya oleh Hakim Mahkamah Tinggi Kuala Lumpur Hakim Datuk Mohamad Zabidin Mohd Diah mengapa ini adalah perlu, Dr McDonald menjawab kerana lebih banyak ujian diperlukan bagi mengenal pasti milik siapa DNA itu (pada 18 *allele).
"Kedua-dua mereka (Seah dan Nor Aidora) tidak menawarkan apa-apa penjelasan mengapa ujian lanjut tidak dilakukan."
Justeru katanya lagi, pakar kimia tempatan itu telah mengabaikan beberapa garis panduan asas makmal untuk memastikan sampel DNA diasingkan sepenuhnya daripada benda asing.
"Dia tidak boleh mengatakan ia milik sesiapa... hanya jika dia melihat melalui mikroskop dan memastikan sampel hanya mempunyai ekstrak sperma asli," katanya lagi.

The DNA of Male Y (supposedly Anwar's) is taken from the towel, toothbrush, water bottle or hair after he was detained in a lock up overnight.
The sperm is Saifool's own.
Both were mixed and then injected into Saifool's anus in the hotel room.
The DNA of the other person must have come from the police officer who did the injection of the mixture into Saifool's anus in the hotel room.
Anwar's DNA did not come his sperm.
It is easy to get Anwar's DNA from other sources like hair, skin, saliva or blood.
Anwar was intentionally kept in a police lockup overnight.
After he left the cell a forensic team went in to sweep the cell.
As easy as ABC..thats what Dr.Seah,
Nor Aidora and the prosecution team thought!

Dr McDonald had previously said the DNA profiles in three sperm samples taken from the complainant Mohd Saiful Bukhari Azlan’s rectum showed “no evidence of degradation”
Dr McDonald said the tests done by the government chemists had failed to show where the DNA samples were taken from.
He had told the High Court that the DNA profile in one high rectal sample was pre-dominantly from “Male Y”, another was pre-dominantly Saiful’s, and the third — from the low rectal area — was inconclusive.
